Hi guys, I really need help with this. How do I permanently move the positions of all of the “drawing substitutions” at the same time? I need to reposition the mouth in my case, so what’s the best way to do this without causing alignment issues?

Currently, the best way to change the artwork all drawings on a given layer is to expose all the drawings in the timeline, then move the artwork using the “apply to all frames” button! 
So:
1- Select the layer
2- expose every drawings from this layer that you want to edit the artwork of
3- Select the Select Tool (black arrow)
4- Go to your Tool Propertie window and select the Apply to All Frames button
5- Move the artwork to the desired spot!
